Cyclic nucleotide phosphodiesterase inhibition increases tyrosine phosphorylation and hyper motility in normal and pathological human spermatozoa Biocell
Yunes,Roberto; Fernández,Pedro; Doncel,Gustavo F.; Acosta,Aníbal A..
Our objective was to determine the effect of phosphodiesterase (PDE) inhibition on: 1) tyrosine phosphorylation of human spermatozoa at the tail level; and 2) sperm motion parameters and hyperactivated motility. The study was conducted with normozoospermic and asthenozoospermic samples incubated under in vitro capacitating conditions. The main outcome measures were computer-assisted sperm motion analysis and fluorescent immunodetection of phosphotyrosine-containing proteins. Pentoxifylline (PTX) was used as PDE inhibitor because of its wide use in the clinic. PTX-treatment significantly increased sperm velocity, hyperactivated motility and tyrosine-phosphorylation, both in normo and asthenozoospermic samples. Tyrosine-phosphorylation of tail proteins was...

Impact of embryo transfer phenotypic records on large-scale beef cattle genetic evaluations R. Bras. Zootec.
Junqueira,Vinícius Silva; Lopes,Paulo Sávio; Resende,Marcos Deon Vilela de; Silva,Fabyano Fonseca e; Lourenço,Daniela Andressa Lino; Yokoo,Marcos Jun Iti; Cardoso,Fernando Flores.
ABSTRACT We aimed to evaluate the impact of using embryo transfer (ET) information on weaning weight estimated breeding values (EBV) and its accuracy. Data from Hereford and Braford cattle, raised under extensive conditions in Southern Brazil, were used. A model that included ET information in addition to maternal (genetic and permanent environmental) effects as a function of foster dams was compared to a model without ET information. Accuracy of both bulls and calves increased due to inclusion of ET records in 0.04 and 0.12 points, respectively. In general, the inclusion of ET records provided a greater amount of phenotypic variance and most accurate EBV for sires and progeny. The results obtained in this study encourage the use of ET phenotypic records...

Sperm handling in aquatic animals for artificial reproduction ArchiMer
Beirão, José; Boulais, Myrina; Gallego, Victor; O’brien, Justine K.; Peixoto, Silvio; Robeck, Todd R.; Cabrita, Elsa.
Artificial reproduction involves collection and handling of gametes in a way that secures their quality and maximizes the fertilization outcome. In addition to initial sperm quality, numerous steps can affect the final result of fertilization, from the sperm collection process until gamete mixing (or co-incubation) when spermatozoon enters or fuses with the oocyte. In this review, we summarize the whole process of sperm handling, from collection until fertilization for fish, penaeid shrimp, bivalve mollusks and marine mammals. To obtain sperm from captive animals, techniques vary widely across taxa, and include stripping by abdominal massage or testis surgical removal in fish, spermatophore collection in penaeid shrimps, gonadal scarification or...
